Hereditary agammaglobulinaemia is caused by loss-of-function variants in genes required for B-cell development, leading to a profound block in B-cell maturation, near-absent circulating B lymphocytes, and a marked reduction of all immunoglobulin classes that manifests as recurrent severe bacterial infections.
